Do any of the big 5 have restrictions to uploading a photo to more than one site? Are there any legal restrictions?

The only restriction that I am aware of is if you are exclusive or if you mark you image as exclusive.  This increases your royalty % but means legally you cant upload to other sites.

I know I can upload to more than one site but the same image can be uploaded to multiple sites. For example: does istockphoto care if I uplaod to them dog1432.jpg and then also uplaod do1432.jpg to shutterstock?

Yes, the same image can be uploaded to multiple sites (as long as you are not exclusive with any one site).
