Title: Change status of image to exclusive

Is it enough to delete an image from the port on other micro stock sites in order to claim exclusivity on one site, or must the image have been uploaded to just the exclusive site?

That is fine. For example if you want to make an image exclusive on dreamstime you click it as exclusive after you deactivate that image from all other rf sites.

That said, that's only true for RF. The rules for exclusive RM may differ in this point.
