I'm ready to start working with stocksubmitter but I've got some newbie questions. I hope you can help me:

1) I already have 100 pictures at Fotolia, mostly vectors. Can I sync/download them somehow with stocksubmitter to avoid double content? Does stocksubmitter recognize already uploaded images even if they were uploaded manually ?

2) Many microstock page like Shutterstock requires to submit work samples before they accept you as contributor. How does stocksubmitter deal with it?

1. Just select images that are already there at Fotolia, click small status icon to the right of the "Fotolia" in the upload/submission panel below and select "Submitted" status for them.
They won't be uploaded to Fotolia if they have "Submitted" status even if you'll select them and Fotolia and click "Upload now".

2. You can use StockSubmitter only if you have passed the exam already.
I'd also recommend to get used with the agency rules before you use StockSubmitter to submit images there because many agencies have specific rules (especially agencies like GraphicRiver) and you can get rejections (or even ban) if you ignore them (especially repeatedly).

Can I tell you some more questions?
3) I ask myself if I can use Stocksubmitter on multiple machines - in this case on my PC and the one of my colleagues PC at the same time? The images are on the intranet using the same drive letter. 
4) Where is Stocksubmitter storing the config informations? In the case I have to migrate them some day to a new PC it would be nice to know which files have to be ported.

3. You can't work on the same files simultaneously because it will lead to the write access conflicts.
You can definitely work on different files in the same folder simultaneously from different copies of StockSubmitter.
There is also problem with releases base synchronizing: if you create and attach releases on the first machine you won't see them on the second and vice versa.
I'm planning to implement releases synchronizing over internet to solve this problem but right now if you need to synchronize releases your only option is to transfer configuration+releases as described in next answer.

4. You can copy all the StockSubmitter's configuration between computers (or back it up and restore later) using menu commands File -> Backup configuration and File -> Restore configuration backup.
